一、测序技术概述

测序技术指用于确定核酸、蛋白质、多糖等生物大分子的一级结构,较为常见的是核酸测序,确定核苷酸在核酸序列中的顺序,包括DNA和RNA测序,简单来说,就是将DNA/RNA分子中碱基ATGC的排列顺序显示出来。

测序技术的发展主要基于两个非常具有里程碑意义的理念:“生命是序列的”和“生命是数据的”,序列是基因组学最基本最重要的数据,也是生命科学领域大数据时代的核心组成部分。目前DNA测序已经从第一代DNA测序技术发展到第三代DNA测序技术,下文就DNA的三代测序技术进行简要说明。

(一)第一代测序技术

一代测序技术以Sanger发明的DNA双脱氧核苷酸末端终止测序法(chain terminator sequencing) [1]和A.M.Maxam和W.Gilbert报道的DNA化学降解测序法(chemical degradation sequencing)为代表[2],但由于化学降解法的程序复杂,后来逐渐被Sanger测序法代替。

基于DNA合成反应的Sanger法,又称为SBS法、末端终止法,于1975年由Sanger提出,并于1977发表第一个完整的生物体基因组序列。其核心原理是由于双脱氧核苷三磷酸 (ddNTP)的2、3位置不含羟基,在DNA合成反应中不能形成磷酸二酯键,因此可以被用来中断DNA合成反应,在4个DNA合成反应体系中分别加入一定比例带有效放射性同位素标记的某种ddNTP(分为:ddATP,ddCTP,ddGTP 和 ddTTP),通过凝胶电泳和放射自显影后,可以根据电泳带的位置确定待测分子的DNA序列[3]。

(二)第二代测序技术

20世纪90年代中期,第二代测序(next-generation sequencing,NGS)技术出现,随着NGS的发展出现了不同的二代测序平台。2005年,出现了第一个商用二代测序平台——454 Roche GS FLX[5],该平台目前已经被淘汰;随后出现了Illumina(2006年)、SOLiD(2007 年)、Ion Torrent(2010 年)和华大基因的 BGISEQ (2013 年)。这些测序平台除了SOLiD是基于连接法测序 (sequence by ligation,SBL)外,其他平台都采用边合成边测序(sequence by synthesis,SBS)技术

Illumina循环SBS法 (cycle SBS)也叫SBRT (Sequencing By Reversible Termination,可逆终止)的核心技术是DNA合成的可逆性末端循环,即3'-OH可逆性的修饰和去修饰。其基本原理是将dNTP的3'-OH以叠氮基团RTG (Reversible Terminating Group,可逆末端基团)进行修饰;将4种碱基分别与不同的荧光分子连接;DNA合成时,RTG能起到类似于ddNTP的作用终止反应;每次合成反应终止并读取信号之后,洗脱RTG和荧光分子,进行下一轮循环[6](如下方视频所示)
